DNA Sequence Analysis of a Monoclonal Antibody AP1 against Human CD61

Abstract
Purpose: This study produced a monoclonal antibody against CD61, a platelet-specific antigen.

Materials and Methods: We extracted RNA using an AP1 hybridoma clone and amplified it through PCR using two specific primers. We made recombinant DNA, and then a sequence of variable heavy and light chains was developed.

Results: The expression of CD61 in platelet was examined. DNA sequences for the CDR regions of heavy and light chain immunoglobulin genes were identified.

Conclusion: DNA sequences of variable heavy and light chain genes for AP1 was developed. Using sequence, we need to select cell line and produce chimeric antibody for the development of therapeutic agent.
